【摘要】 通過IgE誘導大鼠被動皮膚過敏反應(PCA)實驗、細胞毒性試驗、IgE誘導肥大細胞組胺釋放、鈣攝入量實驗、Western blot試驗,來研究樺褐孔菌(Inonotus oblipuus)乙醇提取物EEIO的抗過敏作用。試驗發(fā)現(xiàn),EEIO明顯抑制了大鼠皮膚伊文思藍滲出,對IgE誘導肥大細胞的細胞內(nèi)鈣攝入、組胺以及腫瘤壞死因子-α、白細胞介素6的釋放有劑量依賴抑制作用。表明,樺褐孔菌乙醇提取物具有一定的抗過敏作用。
【Abstract】 We have studied the anti-allergic effects of an orally-administered ethanolic extract of Inonotus obliquus(EEIO) by evaluating anti-DNP IgE-mediated passive cutaneous anaphylaxis(PCA) in rats,EEIO cytotoxicity,histamine release,calcium uptake,and TNF-α and IL-6 secretion by IgE-induced rat peritoneal mast cells(RPMCs) incubated with EEIO.EEIO(1 100 μg/mL) had no adverse effect on RPMC viability under the test conditions,and significantly inhibited Evans blue leakage in antigen-challenged rats sensitized by anti-DNP IgE.EEIO also suppressed antigen-stimulated calcium uptake and histamine release in RPMCs,and Western blot analysis revealed that EEIO inhibited the antigen-stimulated expression of TNF-α and IL-6 in RPMCs in a dose-dependent manner.Our data indicate that EEIO exhibits anti-allergic effects.
