【中文摘要】 以氯磺酸為磺化試劑、吡啶為溶劑半合成制備茯苓多糖硫酸酯(Pachyman sulfate,PS),對(duì)比分析磺化前后產(chǎn)物的特征光譜變化;以自然殺傷細(xì)胞-NK細(xì)胞體外殺傷小鼠淋巴瘤細(xì)胞Yac-1的殺傷率衡量NK細(xì)胞活性,考察PS增強(qiáng)NK細(xì)胞自然殺傷活性。結(jié)果表明,PS重均分子量為1.02×104Da;紫外光譜分析表明PS在260 nm處可見(jiàn)明顯吸收峰;紅外光譜分析表明,PS新出現(xiàn)的在1250 cm-1和810 cm-1處的吸收峰為硫酸酯化的特征峰;13C-NMR譜表明PS單糖殘基上3個(gè)自由羥基均發(fā)生取代反應(yīng);PS顯著提高NK細(xì)胞殺傷Yac-1細(xì)胞的活性(P<0.001),且試驗(yàn)范圍內(nèi)殺傷率隨PS劑量的增加而提高。
【英文摘要】 Pachyman sulfonate(PS)(mol.wt.1.02×104 Da) was synthesized from pachyman(dissolved in pyridine) using chlorosulfonic acid as the sulfonating reagent and the spectral characteristics before and after sulfonation were compared.A distinct absorption peak was observed at 260 nm in the UV-VIS spectrum of PS,and the FT-IR spectrum contained absorption peaks typical of sulfate-substituted glucoside hydroxyl groups at 1 250 cm-1 and 810 cm-1.The 13C-NMR spectrum of PS indicated complete sulfonation at C-6,C-2 and C-4 of the glucoside radical.PS significantly(P<0.001) enhanced the killing activity of NK cells towards Yac-1 cells in vitro in a dose-dependent manner.
