【中文摘要】 用CTAB法提取美味側(cè)耳基因組DNA,利用引物ITS1和ITS4擴(kuò)增其核糖體DNA的ITS片段,凝膠回收試劑盒純化目的片段。通過(guò)PCR產(chǎn)物雙向直接測(cè)序和克隆測(cè)序分別獲得其rDNAITS序列,對(duì)兩種測(cè)序方法所得序列及GenBank中美味側(cè)耳ITS片段的序列進(jìn)行比較分析。結(jié)果顯示,三條序列的差異非常小,兩種測(cè)序方法所得序列僅在兩端出現(xiàn)差異,序列內(nèi)部完全一致。

【英文摘要】 Genomic DNA was obtained from Pleurotus sapidus using CTAB extraction. The nuclear ribosomal ITS was amplified with primers ITS1 and ITS4, and PCR products were purified with a Wizard PCR purification kit. Sequences of ITS obtained by direct sequencing are the same as that of the cloned sequencing except little difference at the both ends of ITS.
