【中文摘要】 采用氯化芐方法提取香菇總DNA,用Gene Pulser Ⅱ電擊系統(tǒng)將香菇DNA導入平菇單核原生質(zhì)體.通過香菇DNA的導入,利用基因互補原理獲得了能出菇的轉(zhuǎn)化子1-1和13-48,并對轉(zhuǎn)化子進行了拮抗試驗、菌絲生長速度分析、酯酶同工酶分析和RAPD分析.
【英文摘要】 Abstract The total DNA of Lentinula edodea isolated by the method of benzyl chloride was introduced into protoplasts of monokaryotic mycelia of P. oatreatua with Bio-Rad Gene Pulser n electroporation system. Optimal electroporation condition was selected in this study. Transfor-mants 1-1 and 13-48 with clamp connection were obtained and formed fruitbodies through principle of genie complementation after introducing L. edodea DNA. While the antagonism reaction test and the mycelium growth rate, esterase isozyme and RAPD analysis of the transformants were also carried out in this study.
