【中文摘要】 脈沖電泳是一種可用于分離20Kb到10Mb大分子量DNA的新型凝膠電泳技術(shù),已應(yīng)用于雙孢蘑菇,香菇、側(cè)耳、草菇等多種食用菌的核型分析及核型多態(tài)性的研究。本文介紹了真菌電泳核型分析的基本操作程序和主要影響因素,并闡述了脈沖電泳在食用菌種群特異性鑒定、染色體作圖和遺傳分析等方面的應(yīng)用前景。
【英文摘要】 Pulsed Field Gel Electrophoresis (PFGE) is a new type technique of gel electrophoresis which can be used to separate large DNA molecules ranging from 20Kb to 10 Mb. It has been applied to the electrophoretic karyotype analysis and karyotype polymorphism research of various fungi such as Agaricus bisporus, Lentinula edodes, Pleurotus spp. and Volvariella volvacea. This article describes the general manipulative procedure and primary effective factors of PFGE, and elaborates the applicable prospect of PFGE in fungal specific identification of species group, chromosome mapping and genetic analysis.
