沒食子酸對金黃色葡萄球菌抑菌活性及機制研究

【摘要】 以金黃色葡萄球菌為供試菌,利用2,3,5-氯化三苯基四氮唑(TTC)染色,細胞膜滲透性測定,SDS-PAGE蛋白譜變化等方法對沒食子酸的抑菌活性及其機制進行研究,進而驗證沒食子酸的藥效作用。實驗結(jié)果顯示,沒食子酸對金黃色葡萄球菌作用其抑菌圈直徑為13mm;其對金黃色葡萄球菌的最低抑菌濃度為0.01mg.mL-1;當藥物作用5h時,實驗組的電導率與對照組相比增加了6.38%;大分子的OD值隨著作用時間的增加而逐漸增大,當培養(yǎng)至8h時,實驗組大分子物質(zhì)吸光值的變化率達到最大,比對照組增加了74.4%。SDS-PAGE蛋白譜變化實驗表明,沒食子酸對金黃色葡萄球菌作用20h后,其總蛋白量沒有明顯變化。上述結(jié)果表明,沒食子酸對金黃色葡萄球菌有較強的抑菌活性,能影響金黃色葡萄球菌細胞膜的通透性。其抑菌機制是通過破壞細菌細胞膜的完整性來實現(xiàn)的。
【Abstract】 To investigate the Antibacterial Activity and Mechanism of Gallic acid on Staphylococcus aureus.The antibacterial activity of Gallic acid was carried out by utilizing oxford cup and 2,3,5-triphenyl tetrazolium chloride(TTC),and the mechanisms of Gallic acid on Staphylococcus aureus were systematically investigated using determining membrane penetrability,the change of SDS-PAGE protein spectra and so on.It is found that Gallic acid has antibacterial activity on Staphylococcus aureus,and the minim...
